The project

The extracellular matrix (ECM) is a critical player for the functioning of multi-cellular organisms and altered functioning of its highly dynamical enzyme-regulated remodeling is associated with major diseases, including cancer metastasis. Yet, quantitative kinetic models of ECM remodeling in realistic conditions are still lacking. In this project we will investigate how the kinetics of selected ECM- remodeling enzymes are influenced by controlled artificial media mimicking some of the features of the ECM, formed by key components that are also substrates for the enzymes, such as collagen, elastin and hyaluronan. To this end, our interdisciplinary consortium, building on preliminary joint work, will perform UV/Vis spectroscopy, capillary electrophoresis and PFG-NMR experiments on systems of increasing complexity. Ultimately, the project will deliver novel kinetic models of enzymatic degradation/remodeling of key ECM components in realistic conditions.
